Classic culture conditions for 129-derived murine embryonic stem cells (mESCs) include culturing in medium containing DMEM, FBS, LIF and co-culturing with primary mouse embryonic fibroblasts (MEFs). Pluripotent mESCs have a very distinct morphology when cultured under these conditions; growing as tightly clustered colonies with smooth phase bright borders. When co-cultured in ES cell media with a mouse embryonic fibroblast (MEF) feeder layer, these stem cell cultures are suitable for electroporation, expansion, or further experimentation.
