Endothelial Cell Adhesion Molecule (ESAM) is a 55 kDa type I transmembrane glycoprotein member of the JAM family of immunoglobulin superfamily molecules. The 390 amino acid Human ESAM contains a 216 amino acid extracellular domain (ECD) with a V-type and a C2-type immunoglobulin (Ig) domain. ESAM is specifically expressed at endothelial tight junctions and on activated platelets and performs homophilic adhesion activity. The adaptor protein membrane-associated guanylate kinase MAGI-1 has been identified as an intracellular binding partner of ESAM. In addition, ESAM at endothelial tight junctions participates in the migration of neutrophils through the vessel wall, possibly by influencing endothelial cell contacts. ESAM-deficient mice were described with lowered angiogenic potential, and accordingly, overexpression of ESAM is closely associated with certain tumor growth and metastasis. ESAM is expressed on endothelial cells, activated platelets and megakaryocytes. The ECD of human and mouse ESAM share 69% amino acid identity.

It plays a critical role in the regulation of cell-surface plasminogen activation in physiological and pathological conditions, and it is also involved in cellular adhesion, the transmission of extracellular signals across the plasma membrane and the subsequent regulation of gene expression. uPAR has been implicated in several biological processes including angiogenesis, monocyte migration, cancer metastasis, trophoblast implantation, and wound healing. Human uPAR is encoded as a 313 amino acid residue polypeptide, excluding a 22 residue signal peptide and shows 60-70% similarity with the murine uPAR amino acid sequence although binding of uPA to uPAR shows strong species specificity.

HSPB11 overexpression protected against etoposide-induced cell death that correlated with a decreased release of mitochondrial Cytochrome C into the cytosol. Inhibition of HSP90 function completely abrogated the protective effect of HSPB11. This data suggests that at least in the case of HSPB11, interaction with other chaperone machines besides HSPA1A may contribute to functional specificity and cellular functioning.
