CD4 is a member of the immunoglobulin superfamily and is implicated as associative recognition element in MHC (Major Histocompatibility Complex) class II-restricted immune response (1). On T-lymphocytes, it defines the helper/inducer subset. The mature 55 kd CD4 protein consists of a 372 amino acid extracellular segment composed of four tandem immunoglobulin-like VJ regions. The CD4 molecule is a major receptor for human immunodeficiency virus (HIV), binding directly to the envelope glycoprotein gp120 on HIV, with the co-receptors being CCR5 or CXCR4. It has been shown that the V-like domains are critical for binding with HIV envelope gp120 (2). This soluble CD4 fragment (sCD4) will work in HIV neutralization assays at levels comparable to previously published results. The minor upper band seen in the Coomassie blue-stained gel is likely to be due to the presence of uncleaved signal sequence.

SLC1A3, also known as EAAT-1 or GLAST, is a membrane-bound protein localized in glial cells and pre-synaptic glutamatergic nerve endings. It transports the excitatory neurotransmitters L-glutamate and D-aspartate, which is essential for terminating the postsynaptic acction of glutamate. Recently, a correlation between expression/function of glial EAAT-1 and tumor proliferation has been reported. The exceptionally rare expression of EAAT-1 in non-neoplastic choroid plexus (CP) compared to choroid plexus tumors (CPT) may distinguishes neoplastic from normal CP. There are a number of splicing variants of SLC1A3, like GLAST1a and GLAST1b, exist due to the exon skipping. It also undergo glycosylation. Variety of bands can be observed in the western blotting assay: 50-55 kDa represents the unglycosylated GLAST1a or GLAST1b, 65-70 kDa correspond to the glycosylated proteins, larger proteins between 90-130 kDa may be the multimers of SLC1A3. The Biosensis Mouse and Rat proNGF Rapid enzyme-linked immunosorbent assay (ELISA) Kit is a sandwich ELISA that allows the quantification of rodent full-length proNGF protein in less than 4 hours in cell culture supernatants, cell lysates and tissue homogenates only if used as directed. This ELISA Kit has also successfully been used on mouse urine samples (Ryu JC et al., 2018). Please refer to the kit protocol for specific use instructions for each substrate application.

This ELISA kit contains a recombinant mouse proNGF standard expressed in E.coli and consists of a pre-coated anti-proNGF capture antibody, a biotinylated anti-proNGF detection antibody and horseradish peroxidase (HRP)-conjugated streptavidin. The addition of a substrate (3,3',5,5'-tetramethylbenzidine, TMB) yields a colored reaction product which is directly proportional to the concentration of proNGF present in samples and protein standards. A proNGF positive control (QC sample) is provided to assure consistent assay performance. This ELISA kit detects rat proNGF due to high degree of homology (96%) with mouse proNGF based on amino acid sequence, and the ability of this kit in detecting proNGF in rat PC12 cell lysates and rat brain tissue homogenate. In the absence of a true rat proNGF standard, results may be expressed as 'mouse proNGF equivalents'. This ELISA kit shows only 20% reactivity with the human form of proNGF and is therefore not suitable to quantify human proNGF. No cross-reactivity was observed with mature mouse NGF and full-length proBDNF when tested in assay buffer.

The antibodies used in this ELISA kit bind epitopes within the pro-domain (capture) and mature domain (detection) of the protein, thus this ELISA assay does not detect the pro-domain peptide. This kit has not been tested for other applications. Sufficient amount of proNGF standard is supplied to allow for spike- and recovery experiments in order to validate this ELISA assay for other sample matrices if required.

Interactions of ICAM-2 and the integrin receptors mediate cell adhesion in a wide range of lymphocyte, monocyte, natural killer cell, and granulocytewith other cells, and play important roles in many adhesion-dependent immune and inflammation responses, such as T cell aggregation, NK-cell cytotoxicity and migration, lymphocyte recirculation, etc. Serum levels of ICAM-2 correlated significantly with the inflammatory and course sequences of trichinosis in mice and had a similar relation with blood eosinophilia. So, estimation of ICAM-2 serum levels may prove useful in diagnosis of trichinosis recent infections, and in monitoring the prognosis and response to treatment.
