Interleukin 6 (IL6) is a multifunctional 24 kD protein originally discovered in the medium of RNA stimulated fibroblastoid cells. It is upregulated by IL1, TNF, PDGF, IFN beta, TNF alpha, NGF, IL17 and downregulated by glucocorticoids IL4, TGF beta. IL6 appears to be directly involved in the responses that occur after infection and cellular injury, and it may prove to be as important as IL1 and TNF alpha in regulating the acute phase response. IL6 has also been implicated in regulating adipose mass. IL6 is reported to be produced by fibroblasts, activated T cells, activated monocytes or macrophages and endothelial cells. It acts upon a variety of cells including fibroblasts, myeloid progenitor cells, T cells, B cells and hepatocytes. In addition, IL6 appears to interact with IL2 in the proliferation of T lymphocytes. IL6 potentiates the proliferative effect of IL3 on multipotential hematopoietic progenitors.

CD95 is a member of the TNF receptor family, which includes TNFR-1, TNFR-2, CD27, CD30 and CD40. Binding of CD95 Ligand to CD95 or crosslinking of CD95 by anti-CD95 monoclonal antibodies leads to apoptosis of CD95 expressing cells. CD95 belongs to a subgroup of family members that have a death domain (DD) which contains 70 amino acids near the carboxyl-terminal region of the molecule. The binding of adaptor molecules to this DD is responsible for transmitting the death signal for apoptosis. Stimulation of CD95 results in aggregation of its DD, leading to the recruitment of FADD and caspase-8 that together with the receptor form the death-inducing signaling complex (DISC).

Together with synaptotagmin and a SNAP, NSF modulates the interaction of SNAP25, VAMP, and syntaxin, in an ATP dependent manner, to form a complex with a sedimentation coefficient of 20 S. This complex may represent an intermediate involved in synaptic vesicle docking and fusion.

The alpha subunit of the IGFI receptor is extracellular while the beta subunit has an extracellular domain, a transmembrane domain and a cytoplasmic tyrosine kinase domain. The IGFI receptor is highly expressed in all cell types and tissues. Essentially all of the biological activities of IGFI and II have been shown to be mediated via the IGFI receptor.

Bassoon does not belong to any known protein families. It has been found in rat and mouse with sequence identity of the two proteins at 96%. The human BASSOON gene has recently been cloned and localized. Bassoon is predicted to contain two double-zinc fingers, three coiled-coil regions, and two polyglutamine domains. The polyglutamine domains in the C-terminus are of interest, since it is known that for some human proteins, such as Huntingtin, abnormal amplification of this region can cause late-onset neurodegeneration. Bassoon is concentrated at sites opposite to postsynaptic densities in synaptic terminals and in cultured neurons, it is found to colocalize with GABA (A) and glutamate (GluR1) receptors.Another presynaptic protein, Piccolo was found to colocalize with Bassoon in cultured hippocampal neurons. MPR cycles and transports newly synthesized lysosomal enzymes between the TGN and late endosomes / early lysosomes. It also cycles and transports extracellular lysosomal enzymes between the plasma membrane and early endosomes via clathrin coated vesicles. The bulk of the MPR protein is in the extracellular/lumenal domain. The entire pool of MPRs cycles between these cellular compartments every 3 hours. The steady state distribution of MPR's is predominantly within late endosomes, fewer in the trans Golgi network and ~5-10 % at the cell surface. In addition to its man6P binding activity, the MPR contains a separate binding site for the type II insulin-like growth factor and is capable of binding both man6P and IGF-II simultaneously. An ~240 kDa soluble, truncated form, representing the extracellular domain of the protein, has also been found circulating in serum and is capable of binding both ligands.
