FACS: Use 10 ?g/ ml is for 10 ?l (10 ^5-6 ) cells. IF: Use at a concentration of 5 ?g/ml (determined using stimulated human peripheral blood mononuclear cells (PBMC) fixed with 2% formaldehyde and permeabilized with 0.1% saponin). IHC: Use 0.5 -5 ?g/ ml (determined using human CXCR4 cultured cells or tissue sections.) Inhib: For neutralization, a working concentration of 2.5-20 ?g/ml of CXCR4 will block 50% of the bioactivity due to 0.1 ?g/ml recombinant human SDF1 in an assay measuring chemotaxis using BaF/ 3 human CXCR4 transfected cells. The ND50 of the antibody is defined as the concentration of antibody resulting in a one-half maximal inhibition of the cell surface CXCR4 mediated SDF1/ PBSF response on a responsive cell line. Not tested in other applications. Optimal dilutions/concentrations should be determined by the end user.
