A propos de cet article
Our AChR is directly purified from the acetylcholine receptor of Torpedo californica.
- Alternative names: Acetylcholine receptor subunit alpha, beta, gamma and delta
- UniProt Number: P02712 (alpha subunit), P02714 (beta subunit), P02718 (gamma subunit), P02710 (delta subunit)
- Protein class: Ion channel, ligand-gated ion channel, receptor
- Organism: Torpedo californica (electric ray)
- Affinity tags: none
- Expression Host: T. californica (native)
- Buffer composition: 137 mM NaCl, 2.7 mM KCl, 10 mM Na2HPO4, 1.8 mM KH2PO4, 5% glycerin; 0,1% Triton
- Purified via: Protein-specific affinity matrix using immobilized agonist ligand followed by dialysis
- Function: After binding acetylcholine, the AChR responds by an extensive change in conformation that affects all subunits and leads to opening of an ion-conducting channel across the plasma membrane.
It is the best example of how customized protein purification beads can be used to isolate a protein without any epitope tags. The protein was purified through an agarose resin coupled to neurotoxin-3 from king cobras, a natural ligand of AChR.
