TCTN2 (tectonic-2 or TECT2) is a type I membrane protein that belongs to the tectonic family, which consists of TCTN1, TCTN2 and TCTN3. Studies in mice suggest that tectonic may be involved in Hedgehog (Hh) signaling, and essential for ciliogenesis. Tectonic is component of the tectonic-like complex, a complex localized at the transition zone of primary cilia and acting as a barrier that prevents diffusion of transmembrane proteins between the cilia and plasma membranes. It exists two spilcing isoforms. The first aberrant transcript introduces 104 base pair from intron 13 and would delete 196 original amino acid, introduce two novel amino acids, and prematurely truncate the 697aa protein at residue number 504. The second transcript lacks exon 14 and would delete 195 original amino acids, introduce four novel amino acids, and prematurely truncate the 697 aa protein at residue number 507. The molecular weight of two isoforms are about 50kDa. We detected the 50kDa and 90kDa protein by immunoprecipitaion, but 50kDa protein may be the splicing isoform or non-specific band.

Signal Regulatory Protein alpha (SIRP alpha) is a monomeric approximately 90 kD type I transmembrane glycoprotein. The 504 amino acid human SIRP alpha contains two Ig-like C1-type domains and one Ig-like V-type domain. SIRP alpha can express in various tissues, mainly on brain and myeloid cells, including macrophages, neutrophils, dendritic and Langerhans cells. It also can detect in neurons, smooth muscle and endothelial cells. SIRPA is an immunoglobulin-like cell surface receptor for CD47. SIRP alpha acts as docking protein and induces translocation of PTPN6, PTPN11 and other binding partners from the cytosol to the plasma membrane. SIRP alpha shows adhesion of cerebellar neurons, neurite outgrowth and glial cell attachment. SIRP alpha engagement generally produces a negative regulatory signal; it may mediate negative regulation of phagocytosis, mast cell activation and dendritic cell activation

BMPs (bone morphogenetic protein) are involved in endochondral bone formation and embryogenesis. BMPR2 encodes a member of the (BMP) receptor family of transmembrane serine/threonine kinases. It is a widely expressed receptor, with high mRNA expression during development in many tissues. Dimer of BMPR2 receptors (70-80 kD) forms a complex with a dimer of type I BMPR1(50-55 kD) in signal transduction. VPS41 (vacuolar protein sorting 41) is the human ortholog of yeast Vps41 protein. Yeast Vps41 is a subunit of AP3 (heterotetrameric adaptor protein complex 3) and HOPS (homotypic fusion and protein transport) complexes and is involved in trafficking of proteins from the late Golgi to the vacuole. In mammalian cells, Vps41 was found to be a member of HOPS complex, mediating the trafficking of proteins to lysosomes . It has been reported that human VPS41 has a HOPS-independent function in the LAMP carrier pathway and also participates in lysosomal maturation, probably as part of the HOPS complex .

Human signal recognition particle 54kDa. The signal recognition particle (SRP) is an oligomeric complex that mediates targeting and insertion of the signal sequence of exported proteins into the membrane of the endoplasmic reticulum. SRP consists of a 7S RNA and six protein subunits. 54 kD protein (SRP54) is one of these subunits. It is a GTP-binding protein that interacts with the signal sequence when it emerges from the ribosome. The 54K subunit of the signal recognition particle has a two domain structure: the G-domain that binds GTP and the M-domain that binds the 7s RNA and also binds the signal sequence. The N-terminal 300 residues of SRP54 include the GTP-binding site (G-domain) and are evolutionary related to similar domains in other proteins.
