Anti-CD62L Rat Monoclonal Antibody (FITC (Fluorescein Isothiocyanate)) [clone: MEL-14]

Supplier: BIOGEMS INTERNATIONAL INC.

The MEL-14 monoclonal antibody specifically reacts with L- selectin (CD62L), a receptor with lectin-like and Epidermal Growth Factor-like domains. The weight of the CD62L molecules depend on their origin, 74 kDa on lymphocytes or 95 kDa on neutrophils. In the mouse organism, CD62L can be expressed by most thymocytes, on subsets of B and T lymphocytes, monocytes, eosinophils, and neutrophils. The L-selectin binds sulfated, fucosylated, and glycosylated glycoproteins (MadCAM-1, GLYCAM-1, and CD 34). It mediates the migration of lymphocytes to the site of inflammation and their return to the peripheral lymphoid tissues and to the HEV (high endothelial venules). In vitro, L-selectin inhibits this binding and the lymphocyte extravasation into peripheral lymph nodes. After cell activation, it is quickly shed by neutrophils and lymphocytes.

Anti-CD8a Mouse Monoclonal Antibody (PE (Phycoerythrin)) [clone: RPA-T8]

Supplier: BIOGEMS INTERNATIONAL INC.

The RPA-T8 monoclonal antibody specifically reacts with the human CD8a molecule, a 32 kDa cell surface receptor expressed either as a heterodimer (CD8 α/β) or as a homodimer (CD8 α/α) on the majority of thymocytes, a subpopulation of mature T cells, and natural killer cells. CD8 interacts with the major histocompatibility complex class I (MHC class I) molecules on antigen-presenting cells or epithelial cells. The RPA-T8 antibody reacts with 13-48% of peripheral lymphocytes, 80% of thymocytes, and a subset of natural killer cells. RPA-T8, OKT8, and HIT8a antibodies do not compete with each other for binding to peripheral leukocytes, meaning that that they do not recognize the same epitope or block each other by steric hindrance.

Anti-CD40 Mouse Monoclonal Antibody (APC (Allophycocyanin)) [clone: 5C3]

Supplier: BIOGEMS INTERNATIONAL INC.

The 5C3 monoclonal antibody specifically reacts with human CD40, a 48kDA type I glycoprotein. CD40 is part of the TNFR family and expressed on a subset of T cells, B cells, dendritic cells, and macrophages. Anti-TCRB Armenian Hamster Monoclonal Antibody (APC (Allophycocyanin)) [clone: H57-597]

Supplier: BIOGEMS INTERNATIONAL INC.

The H57-597 monoclonal antibody is specific for the beta chain of the mouse T cell Receptor (TCR). The crosslinking induces activation and proliferation of T cells, as a plate-bound or a soluble H57-597, or the plate-bound antibody can induce apoptosis, based on assay conditions. The beta chain of the TCR can combine with the alpha chain of the receptor to produce the alpha-beta TCR, which is expressed by the NKT cells, by the NK1.1+ thymocytes and most of the T cells. The beta chain does not react with the gamma-delta TCR-bearing cells, expressed by a small number of T cells. This antibody can be used as a phenotypic marker for the TCR beta expressing cells or for the functional purpose of TCR-mediated cell activation or apoptosis.

Anti-IL-13 Mouse Monoclonal Antibody [clone: PVM13-1]

Supplier: BIOGEMS INTERNATIONAL INC.

The PVM13-1 monoclonal antibody bings to human IL-13. IL-13 is an immunoregulatory cytokine produced primarily by activated Th2 cells, and also by mast cells and NK cells. Targeted deletion of IL-13 in mice resulted in impaired Th2 cell development and indicated an important role for IL-13 in the expulsion of gastrointestinal parasites. IL-13 exerts anti-inflammatory effects on monocytes and macrophages and it inhibits the expression of inflammatory cytokines such as IL-1β, TNF-α, IL-6 and IL-8. IL-13 has also been shown to enhance B cell proliferation and to induce isotype switching, resulting in increased production of IgE. Blocking of IL-13 activity inhibits the pathophysiology of asthma. The PVM13-1 antibody is reported to be a neutralizing antibody.

The alpha’ chain is the truncated form of alpha chain, encoded by the same CD8a gene. In CD8a, the alpha and alpha’ chains form heterodimers with CD8b (the beta chains) or homodimers (alpha-alpha), which occur as receptors on the surface of the majority of thymocytes. A subpopulation of mature T lymphocytes expresses the CD8 alpha beta (alpha beta TCR T cells), and a subpopulation of intestinal intraepithelial lymphocytes and dendritic cells express CD8a without CD8b.CD8 interacts with the mouse major histocompatibility complex class I (MHC class I) molecules on antigen-presenting cells or epithelial cells. Its function seems to be to attenuate the CD8-mediated signal for the stimulation of intrathymic T-cell maturation. The 53-6.7 antibody is useful for depleting CD8+ peripheral T lymphocytes. It cross reacts with the alpha- and alpha’-like polypeptides on some thymic and peripheral lymphocytes.
