KLRG1 can regulate, in an inhibitory fashion, the development and effector functions of NK cells, and is often cited as a senescence or terminal differentiation marker for T cells. Ligands for KLRG1 include members of the cadherin family of adhesion molecules, specifically N-Cadherin, E-Cadherin, and R-Cadherin. These interactions may induce bidirectional, immunosuppressive signaling in both KLRG- and Cadherin-expressing cells. A more recently identified role for KLRG1-Cadherin signaling in tissue organization, e.g. in cardiac angiogenesis, expands the function of these interactions beyond immunosuppression of immune cells. The 3G3 antibody reacts with mouse Foxp3, a 50-55 kDa transcription factor which is a central regulator of T cell activity and is critical for the development and function of regulatory T cells (Tregs). Foxp3 is expressed at constitutively high levels in Treg cells, which are further identified as being CD4+ CD25+. In resting conventional T cells (CD4+ CD25-) Foxp3 expression is restricted, and upon TCR activation is expressed only transiently and in a small proportion of cells. However, the growth factor TGF-beta has been shown to induce expression of Foxp3 in naïve T cells, driving their development into Foxp3+ Tregs, which are called “induced” or “adaptive” Tregs. These cells are phenotypically similar to so-called “natural” Tregs (CD4+ CD25high Foxp3+) which originate in the thymus and comprise the majority of Treg cells. Tregs are critical for maintaining peripheral tolerance and are implicated in the development of autoimmunity. It is important to review the literature in choosing an antibody for the Foxp3 antigen in flow cytometry, as the potential for high background or non-specific staining may be observed. The 3G3 antibody may be used for intracellular detection of Foxp3 in cells from mouse and Rhesus macaque.

This protein forms homodimers which associate with microtubules in the cytoplasm. The protein is likely involved in the formation of multiprotein structures acting as anchor points to microtubules. Mutations in this gene have been associated with the X-linked form of Opitz syndrome, which is characterized by midline abnormalities such as cleft lip, laryngeal cleft, heart defects, hypospadias, and agenesis of the corpus callosum. This gene was also the first example of a gene subject to X inactivation in human while escaping it in mouse. Several different transcript variants are generated by alternate splicing; however, the full length nature of two variants has not been determined.

In response to adverse changes in their environment, cells from many organisms increase the expression of a class of proteins referred to as heat shock or stress proteins. One class of stress proteins, termed the ""small heat shock proteins"" is comprised of a diverse group of proteins from ~15 to >30kDa, with a single form found in yeast and human cells and multiple related forms found in higher plants. A direct relationship between elevated stress protein expression and neoplasia has been suggested from studies on estrogen-induced proteins in breast cancer cells. In addition, Hsp27 exhibits rapid increased phosphorylation in response to various mitogens, tumor promoters (e.g. phorbol esters) and calcium ionophores. The expression of a 24kDa protein was shown to correlate with steroid hormone receptors in cell lines, organs , and tumors. The 24kDa estrogen-induced protein was found to be expressed at high levels in carcinoma of the breast. Another study has suggested that the levels of Hsp27 correlate with endometrial adenocarcinomas. Interestingly, the regulation of human Hsp27 by estrogens and heat shock is reminiscent of the ecdysteroid and heat shock activation of the drosophila Hsp20 genes.
