ATG13 Antibody: Autophagy, the process of bulk degradation of cellular proteins through an autophagosomic-lysosomal pathway is important for normal growth control and may be defective in tumor cells. It is involved in the preservation of cellular nutrients under starvation conditions as well as the normal turnover of cytosolic components. This process is negatively regulated by TOR (Target of rapamycin) through phosphorylation of autophagy protein ATG1. ATG13 forms a complex with ULK1 and ULK2, the mammalian homologs of ATG1, and with FIP200. This complex is a target of TOR phosphorylation under normal conditions; inhibition of TOR by rapamycin or leucine deprivation leads to dephosphorylation of ATG13, ULK1 and ULK2, which then leads to autophagy. Knockdown of ATG13 inhibits autophagosome formation.

THEM2 Antibody: THEM2, also known as ACOT13, belongs to the hotdog-fold superfamily and possesses thioesterase activity, with greater activity observed with longer chain acyl-CoAs such as myristoyl- and palmitoyl-CoA. THEM2 is highly expressed in several tissues such as heart, kidney, liver and brain and has been shown to be co-localized with beta-tubulin on microtubules. THEM2 interacts with StarD, a protein that plays a key role in fatty acid metabolism, and the addition of StarD to THEM2 increased its catalytic activity, suggesting that THEM2 plays a significant role in the metabolism of fatty acids. At least two isoforms of THEM2 are known to exist.

APRIL, along with another TNF family member, BAFF, compete for two receptors, TACI and BCMA. APRIL has the ability to stimulate proliferation of various tumor cell lines including Jurkat T cells and MCF-7 carcinoma cells. Like BAFF, APRIL also stimulates the proliferation of B and T cells. The human APRIL gene codes for at least four alternatively spliced transcriptional variants, which give rise to different isoforms of the APRIL precursor protein. All isoforms can be cleaved by the protease, furin, to release a soluble C-terminal fragment, which comprises the TNF like receptor binding of the APRIL precursor. Recombinant murine APRIL is a soluble 21.9 kDa protein, consisting of 192 amino acid residues.
